R., Chauvin, A., Pascaud, F., Kellenberger, S. & Farmer, E. E. GLUTAMATE RECEPTOR-LIKE genes mediate leaf-to-leaf wound signalling. Nature 500, 422-426 (2013) でした.傷害ストレスシグナルの葉から葉への伝達にはグルタミン酸受容体様タンパク質を介した電気的シグナルが関与していることを報告した論文です.

化学生態学実験 III,研究室セミナー,輪読
しばらくブログ更新をさぼってしまいました.長谷川担当の化学生態学実験 III は 11/22 で全日程を終了しました.GC/MS で籾殻抽出物中のモミラクトン A を検出する実験が一回目はどの班でも検出できないという問題がありましたが,濃度を上げたところ,全ての班が検出することができました.それ以外は特に大きなトラブルもなく,順調に終了しました.TA の大学院生の堀江君と濱名君のおかげです.レポート提出〆切は今週末 12/6 です.

化学生態学実験 III,研究室セミナー,今週の輪読
今週の月曜日から長谷川が担当する資源生物科学科三年生向けの学生実験「化学生態学実験 III」が始まりました.今年の受講者は 27 名です.今日で前半を終了しましたが,ここまではまだ結果が何も出ていない状態なので,うまくいっているかどうかははっきりしませんが,見たところまあまあ順調に進んでいるようです.

送風乾燥機修理 & 今週の輪読
研究室では長谷川が学生の頃から 20 年以上使っているアドバンテックの送風乾燥機があるのですが,しばらく前からファンがイオンを出すようになっていて,9 月末には時折ファンが停止するような状態になってしまいました.古い機器なので,もう修理は無理かもしれないと半分諦めながらメーカーに問い合わせてみましたところ,ファンモーターの故障ならまだ修理可能ということで,引き取り修理をお願いしていました.その乾燥機が木曜日に一ヶ月ぶりに戻ってきました.戻ってきた乾燥機はファンの音が非常に静かになっていて,周りがうるさい実験室では動いているのが分からないぐらいになりました.ここのところ,製氷機,冷却水循環装置,送風乾燥機と立て続けに研究室のベテラン機器(いずれも 20 年以上使用)が不調に陥っていますが,それぞれ何とか使い続けることができています.

研究室ではエバポレーターの冷却水を循環させるために東京理化の冷却水循環装置が常時 3 台動いています.しかし,その中の一台はもう 20 年以上使っている者で,かなり老朽化していました.この古い冷却水循環装置が 10 月初めから電源が入らなくなって使えなくなってしまいました.さすがにもう修理はできないかと思ったのですが,念のために東京理化に問い合わせたところ,ブレーカーを交換すれば直るかもしれないということになり,今日エンジニアの方に修理してもらいました.予想通りブレーカーの交換で直ったのですが,循環ポンプがかなり危険な状態(漏電スレスレ)なので,もう使わない方が良いと言われてしまいました.さすがに,この循環ポンプはもう修理できないとのことでした.しかし,冷却機の方は生きているので,普段は使っていない開放系用冷却水循環装置(冷却能力が弱い)を循環ポンプとして使うことにして,2 台を繋いで当面は使い続けることにしました.場所をとるのが難点ですが,とりあえず順調に動いています.予算に余裕があれば新品購入したいところですが,今年度はまだ半年近くあるので,現時点での新品購入は難しいのです.エンジニアに来てもらったついでに,まだ 2, 3 年しか使っていない新しい冷却水循環装置も不調だったので,これも見てもらって快調になりました.

研究室セミナー
先週は台風接近で中止になったので,二週間ぶりの研究室セミナーでした.今日の文献紹介の担当は化学生態学研究室修士二年の吉田さんで,紹介した論文は Xia, J., Yamaji, N. & Ma, J. F. A plasma membrane-localized small peptide is involved in rice aluminum tolerance. Plant J 76, 345-355 (2013) でした.イネのアルミニウム耐性に関与する原形質膜局在性ペプチドについての論文です.

市立銚子高校模擬授業,研究室旅行,中間発表 & 先週・今週の輪読
先週はブログ更新をさぼってしまいましたが,この二週間はいろいろな出来事がありましたので,まとめて書きます.
9/26(木)は長谷川が銚子市立銚子高等学校での模擬授業を実施してきました.二年生が対象で「はじめての農薬学」というタイトルで農薬の定義や安全性などについての 60 分の講義を 2 回行いました.参加者は計 30 名程度でした.この日は台風の接近の影響で強風だったので,成田線のダイヤが大幅に乱れて,特に帰りは大変でした.
9/27(金)は生物制御化学研究室・化学生態学研究室合同での研究室旅行が開催されました.旅行といっても近場の日帰りレクリエーションでした.午前中は石岡のミッドクリークフィッシングエリアという釣り堀でルアーを使ったニジマス釣りをしました.ルアーで釣るのはなかなか大変で,しかも竿が 4 本だけで全員で同時には釣ることができなかったため,釣果は戸嶋先生 2,鈴木先生 1,堀江君 1 の合計 4 匹とやや寂しいものでした.釣った魚は焼いて 3 人で一匹の割合で食べました.これだけでは食事にならなかったので,併設のレストハウスで昼食を取りました.釣りの後は土浦のパニックボウルつくばでボーリングを 2 ゲーム行いました.これも戸嶋先生が圧倒的に一番良いスコアでした.最後は阿見に帰ってきて,食いしん坊炎で飲み会でした.
10/2(水)には研究室の中間発表会が開催されました.なかなか研究計画通りには進んでいませんが,卒論や修論をまとめるには残り時間はせいぜい 4 ヶ月程度とあまり残っているわけではないので,一層の奮起を期待しています.

病原細菌に対する抵抗性反応として起こる気孔の閉孔にはアブシジン酸には依存しないオキシリピンのシグナル伝達経路が関与していることを示した報告です.

本日の研究室セミナーの文献紹介の担当は化学生態学研究室四年生の鈴木君で,紹介した論文は Nobusawa, T. et al. Synthesis of very-long-chain fatty acids in the epidermis controls plant organ growth by restricting cell proliferation. PLoS Biology 11, e1001531 (2013) でした. 植物表皮の超長鎖脂肪酸生合成が植物の生長を制限していることを報告した論文です.

2011 年 3 月に修士課程を修了した下村伸君が筆頭著者の論文が Biosci. Biotechnol. Biochem. 誌(日本農芸化学会の英文誌)のサイトでオンライン公開されました.

植物の花芽の形成は日長などの環境要因によって厳密に制御されています.花咲か爺さんの昔話にあるように,花芽形成を人間の意のままに自由に制御することができれば,その技術は農業生産や育種,園芸の現場などで大変役に立つはずです.
この論文は,アオウキクサから単離された花芽形成促進物質 KODA 類縁体の立体選択的合成を報告したものです.KODA は資生堂の横山らによって発見された化合物(こちらを参照)ですが,そのまま植物生長調節剤として用いるには不安定な化合物であるため,本研究室ではシクロプロパン環を導入した化学的に安定な類縁体を設計し,その合成方法について研究を行ってきました.本論文で報告した類縁体が花芽形成促進活性を有するかは今後の研究によって明らかにしなければいけませんが,有効な花芽形成促進物質となることが期待されています.
この論文は下村君だけでなく,今年修了した大山秀芳君と 2008 年修了の中野恭平君たちの修士論文研究成果の一部をまとめたものです.

Resistance of Arabidopsis thaliana to the green peach aphid, Myzus persicae, involves camalexin and is regulated by microRNAs. New Phytol. 198, 1178-1190 (2013) でした. モモアカアブラムシに対するシロイヌナズナの抵抗性にフィトアレキシンであるカマレキシンが関与していて,その制御にはマイクロ RNA が関わっていることを報告した論文です.

今週から研究室セミナーで文献紹介が始まりました.また,先週は就職活動等で発表できなかった四年生 3 名の年間計画発表も行われました.
本日の研究室セミナーの文献紹介の担当は化学生態学研究室修士二年生の吉田さんで,紹介した論文は Wright, G. A. et al. Caffeine in floral nectar enhances a pollinator’s memory of reward. Science 339, 1202-1204 (2013) でした.コーヒーノキや柑橘類の花の蜜に含まれるカフェインがミツバチの匂いの記憶を強化するという内容の論文です.

年間計画発表会 & 先週の輪読
先週の水曜日には生物制御化学研究室と化学生態学研究室合同での年間計画発表会が開催されました.今年度は両研究室合計で 10 名の学生さんがいるのですが,昨年のメンバーで残っているのは 2 名だけで 8 名が新しいメンバーとなりましたので,皆まだ研究が始まったばかりです.研究は大抵の場合は計画通りには進まないものですが,うまくいかないところをうまくいくようにしたり,予想外の結果から面白い成果が得られたりするのが研究なので,10 ヶ月後の修論・卒論発表会では良い成果が発表されることを期待しています.

先月,修士課程を修了した井上靖乃さんの論文がBiosci. Biotechnol. Biochem. 誌にオンライン公開されました.

紫外線照射イネ葉から,新規ジテルペン化合物 ent-10-oxodepressin を単離・同定し,この化合物がイネいもち病菌接種によっても蓄積が誘導され,いもち病菌胞子に対する抗菌活性を有することも示して,イネの新規フィトアレキシンであることを明らかにした論文です.この化合物は従来知られていたイネのジテルペン系フィトアレキシンとは全く異なるカスバン骨格を持つ化合物でした.カスバン型ジテルペンとしては,トウダイグサ科植物と Sinularia 属軟質サンゴ以外の生物からは初めての発見です.また,イネのフィトアレキシンとしては 1997 年のファイトカサン E の発見の報告依頼 16 年ぶり 16 個目の発見となりました.
この研究は井上さんの修士論文研究の一部ですが,卒業・修了生の阪井美紀さん,姚群さん,谷本洋輔君の卒業論文・修士論文研究が引き継がれてまとめられたものです.

研究室ウェブサイトに新たなコンテンツとして「イネのフィトアレキシン」というページを作りました.イネフィトアレキシンに関する色々な情報を掲載していこうと考えていますが,まずは関連する文献リストを最初のコンテンツとして公開しました.このリストもまだ不完全ですが,順次アップデートしていく予定です.

卒業式 & 日本農芸化学会 2013 年度大会
3/26(火)に茨城県武道館で茨城大学の卒業式が挙行されました.今年度は生物制御化学研究室からは修士 2 名,学士 2 名の学生さんたちが卒業することになりました.卒業論文や修士論文研究に取り組んだ過程で培った(はずの)力を生かして,社会へ出ても活躍されることを期待しています.

謝恩会 & 今週の輪読
今年の茨城大学卒業式は 3/26(火)に開催予定なのですが,資源生物科学科の多くの教員と学生が参加する日本農芸化学会(3/24-27)と日程が重なってしまったため,謝恩会は 3/18(月)につくばのホテルグランド東雲で開催されました.今年はうちの研究室の学生さんたちが当番で幹事になっていたので,準備や司会で大変だったようですが,無事に楽しい謝恩会が開催できたと思います.

新しい遠心エバポレーター & 今週の輪読
試料溶液の濃縮操作ではロータリーエバポレーターが多用されますが,少量試料を同時に多数濃縮する必要がある場合には遠心エバポレーターが便利です.研究室では 15 年以上使っている東京理化の遠心エバポレーターがあったのですが,やや回転が不安定で昨年は一回壊れて修理に出したりしていました.幸い予算の都合が付いたので,東京理化の遠心エバポレーター CVE-3100 を新たに購入することができました.同時に同じく 15 年以上使っていたバキューブランドのダイヤフラム真空ポンプ(スイッチが壊れていて,ときどき電源オンできなくなっていた)もビュッヒの V-700 に置き換えることにしました.今週から使い始めましたが,新しい遠心エバポレーターは回転音が非常に回転音が静かです.

新しい真空ポンプ
今日は新しいビュッヒのダイヤフラム真空ポンプの設置作業がありました.研究室では有機溶媒中に溶けている化合物を取り出すために,頻繁に濃縮操作が行われます.そのためにはロータリーエバポレーターや遠心エバポレーターという機器が用いられるのですが,これらは大気圧よりも圧力を低くすることによって液体の沸点を低下させて効率よく溶媒を蒸発させるものです.これらの機器を用いるためには真空ポンプが必要となるのですが,真空ポンプで減圧するときに圧力を低くしすぎると突沸という急激に沸騰が起こる状態になり,困ったことになります.従来は非常に難しいコックの操作をして突沸が起こらないように対応していたのですが,新しい真空ポンプは圧力を精密に制御できるコントローラーが付いているので,エバポレーター使用中の突沸がかなり防げるのではないかと期待しています.

超音波洗浄器更新
研究室で私が学生だった頃から二十年以上使っている超音波洗浄器(シャープ製)があるのですが,一昨年ぐらいからタイマーが壊れてしまいました.超音波自体には特に問題がなかったため,そのまま使い続けていたのですが,予算の都合が付いたので,新しい超音波洗浄器を購入しました.アズワンで売っているものを購入したのですが,従来のものに比べると洗浄槽が浅いのが難点です.また,超音波の出力もだいぶ弱いような気がします.安価なものを買ったので,仕方がないところなのかもしれません.古い方の洗浄器もまだ使えないわけではないので,サブとしてしばらくは使い続ける予定です.

20 年以上使っている超音波洗浄器.さびてぼろぼろだけれど,タイマー以外は問題なし.

卒業論文発表会
今日と明日は資源生物科学科の卒業論文発表会が開催されています.生物制御化学研究室の四年生 2 名は今日の午後の順番だったので,すでに無事に発表を終えました.平石由貴さんが「イネフィトアレキシン モミラクトンAのいもち病菌による代謝」,渡邊峰花さんが「カビ毒ニバレノールの簡易スクリーニング手法の開発 ―ニバレノール抗原および蛍光トレーサーの合成研究―」というタイトルでの発表でした.二人とも発表自体はまあまあの出来だったと思いますが,どちらも研究内容が目標到達まであと一歩(数歩?)というところで時間切れになってしまったのが残念なところでした.卒論発表会は他の研究室の学生さんの発表が明日も続きます.また,明日は卒業論文の提出〆切日にもなっていますので,もうひと頑張りが必要です.

修論・卒論,期末試験,今週の輪読
いよいよ修士論文や卒業論文が大詰めの時期になってきました.先週末の 2/1 が修士論文の提出〆切日で生物制御化学研究室の学生さんたちの修士論文予備審査会は今週の水曜日 2/6 の午後に開催されました.2/7 には修士論文と卒業論文の発表練習会が化学生態学研究室と生物制御化学研究室合同で開催され,来週の本番の発表に向けて多くの要改善点が指摘されました.修士論文発表会は来週火曜日,卒業論文発表会は木,金曜日に開催予定です.また,今日で期末試験が終わりましたので,研究室に所属していない学生さんは明日から二ヶ月間の長い春休みに入ります.四年生と修士二年生の学生はもうひと頑張りです.

研究室セミナー
本日の研究室セミナーの文献紹介の担当は化学生態学研究室四年生の貫井君で,紹介した論文は Behie SW, Zelisko PM, Bidochka MJ (2012) Endophytic insect-parasitic fungi translocate nitrogen directly from insects to plants. Science 336: 1576–1577 でした.昆虫の病原菌でもあるエンドファイトが植物へ窒素を供給しているという内容の論文です.
